[h=2]Introduction[/thanks]Recent advances in next generation sequencing (NGS) technologies have made sequence acquisition more affordable, leading to its use in settings such as the clinic, contract research organizations and small research groups. Such facilities are adopting NGS partly because of the availability of new bench top platforms such as the Ion Torrent PGM from Life Technologies and the MiSeq from Illumina have. In fact, groups that previously used to outsource sequencing to large science facilities can now bring their projects in-house.
NGS is quickly becoming increasingly popular, despite the fact that most liquid handling workstations are validated to complete the library preparation protocols. As NGS requires a high number of sample manipulations, liquid handling needs to be automated for results to be consistent and accurate.
[h=2]VERSA Mini NGLP[/thanks]The VERSA Mini NGLP automated liquid handling workstation offers delicate sample handling, making it suitable for NGS library preparation protocols. The combination of high resolution stepper motors and a single channel, dual syringe pipette manifold allows precise liquid handling at variable speeds across a large volume range.
Sample cleanups, incubations and reaction setup are performed in a UV lamp, HEPA filtered hood. In addition, flexible software provides excellent control over the mechanics of liquid handling. Aspiration and dispense processes can be set at different speeds, depending on the specific operation and new protocols can easily be automated. Furthermore, the number of samples to be processed can easily be updated by users.
[h=2]NEXTflex Library Prep Kits[/thanks]NEXTflex kits from Bioo Scientific are specifically designed for the preparation of single, paired-end and multiplexed genomic, mRNA, small RNA, ChIP and reduced representative bisulfite sequencing libraries. The kits use improved enzymatic master mixes and flexible multiplexing options. One such example is the NEXTflex enhanced adapter ligation technology. The kits are compatible with high-throughput liquid handling automation.
As sequencing throughput improves, users are able to pool a greater number of samples into one sequencing reaction. In order to accommodate this, NEXTflex DNA barcodes can be used to multiplex or tag samples in order to monitor cross contamination with up to 96 indices. This considerably reduces the costs associated with sequencing, as well as enabling measurement of base error rate, cross genomic studies, time course studies and drug induced experiments. Available in 96 well plates, the NEXTflex DNA barcodes allow high throughput automation.
[h=2]VERSA and NEXTflex[/thanks]Labs with multiple NGS technologies can easily automate different library preparation protocols using one reagent provider on the same automated platform. At present, most automation options for library preparation are closed systems that cannot accommodate a wide range of reagent kits.
